The overall goal of this proposal is to determine the role that a novel cell surface protease expressed in human lung cancer cells plays in the development of lung cancer metastasis. We have identified a cell surface serine protease, SPP, that is expressed in the majority of human lung cancer cell lines and in lung cancer tissue. We have also found that SPP is a serine protease with limited tissue distribution. Our preliminary data show that SPP is a matrix metalloproteinase (MMP) and capable of digesting the extracellular matrix. The discovery of a novel human MMP in lung cancer cells suggests that this enzyme may play an important role in the malignant transformation and tumor progression of lung cancer. The specific aims are: 1) To determine if SPP and the enzymatic activity of SPP play an important role in the development and progression of human lung cancer, 2) To determine if SPP has the ability to stimulate the growth of human lung cancer cells by secreting growth factors into the extracellular space, and 3) To determine the tissue distribution and biological function of SPP in the normal lung. We will utilize lung cancer cells that overexpress SPP as a model to determine the contribution of SPP in the malignant transformation of lung epithelial cells. We will also use a SPP-deficient cell line to determine if the loss of SPP affects lung tumorigenesis in a xenograft mouse model. The novel structure of SPP allows us to screen a library of antibodies against this enzyme. We will use these antibodies to examine the expression of SPP in lung cancer and to identify the biological functions of SPP.Q:
